Absorption: The prodrug, diloxanide furoate, is metabolized in the gastrointestinal tract to release the active drug, diloxanide. In parental form, diloxanide is 90% orally bioavailable.

Metabolism: Diloxanide furoate is metabolized to furoic acid and active drug, diloxanide. This active drug is 99% glucuronidated in the liver and 1% is left as free drug in the blood. Furoic acid is metabolized by intestinal esterases.

Excretion: Although 99% of the active drug is metabolized by the liver, 90% of that is excreted in the urine. The other 10% is excreted in the feces.[1]
